Real Time RT-PCR developed in recent years represents a useful tool for quantifying specific target DNA sequences. Compared to relative quantification, absolute quantification has the advantage of determining the absolute copy number of a given target, such as pathogen or plasmid DNA in vivo. Absolute quantification determines the input copy number of the transcript of interest, usually by relating the PCR signal to a standard curve. Plasmids are the preferred standards for absolute quantification. The most convenient way to create a DNA standard is to clone a PCR product into a standard vector. Advantages of this method are that its identity can be verified by sequencing, and the DNA can easily be quantified by spectrophotometry. Plasmid standards should be linearized upstream or downstream of the target sequence, rather than using supercoiled plasmid for amplification. This is because the amplification efficiency of a linearized plasmid often differs from that of the supercoiled conformation and more closely simulates the amplification efficiency of genomic DNA or cDNA.

For our customers, we provide plasmid standards based on the sequence given.

For each standard we need the following items from customer end:

  • 50 mL of fresh positive PCR product using the primer/probe. Which we will purify and try to insert it in the plasmid.
  • Primers, probe (if applicable) and the amplicon sequence. We require this so that if the first approach fails then we will synthesize the amplicon and try to insert in the plasmid.
  • Primers and probe (if applicable). We require this as, after successful synthesis we check its functionality in real time PCR, which is a part of our QC. After this particular QC we send the QC report (the standard curve and the efficiency data) along with the product, so that customer can expect the same result with the product.
